RNA polymerase would begin transcription unsystematically on the DNA. Additionally, an in vitro transcription process changes a bacterial gene however it terminates inefficiently. Furthermore, if a specific sigma subunit of bacterial RNA polymerase is mutated, RNA polymerase would not be successful in initiating transcription at the promoter specifically to the sigma subunit.
